Hidden in Resident Evil Requiem's New Game Plus mode is a multi-step puzzle that ties together "The Girl" antagonist, the Severed Hand, Marie's Doll, and the Sun Quartz Box from Grace's chapters. Solving it unlocks a secret ending dialogue and special trophy. This guide covers every step and the RNA-to-symbol decoding.

Why this puzzle exists
"The Girl" is a recurring antagonist throughout Grace's chapters — a child-like figure with bandaged hands who appears in dreams, hallucinations, and key story moments. After the main credits roll on a first playthrough, players who pay attention to her dialogue notice a strange chant: "Let's play. GGC-AAG-AUA-ACG-UGU-CAU."
This is RNA notation — the four letters G/A/U/C represent the four ribonucleotides (Guanine, Adenine, Uracil, Cytosine). Combined with the Severed Hand item Grace can find in NG+, these letters decode to a Sun-Moon-Star symbol sequence that opens a hidden puzzle in the Lead Researcher's Office.

💡 How to discover this mapping: Three Files in the Care Center reveal the encoding through ARK research notes. They describe Cytosine and Guanine as "the bonded pair" — both mapped to Sun. Adenine pairs with Uracil — Moon and Star respectively. Reading all three Files unlocks an in-game File entry that confirms the mapping.

What the puzzle reveals
The cutscene reveals Marie's connection to "The Girl" — they are sisters separated by Capcom's bioweapons program. Marie's Doll was originally The Girl's, lost when she was taken by Umbrella researchers years before the events of RE9.
The puzzle's solution implies that Marie chose to remain in the Care Center as a "guardian" of The Girl's spirit, refusing to leave even when offered escape. This thread closes a major emotional arc that's only hinted at in the main story.
