Hidden in Resident Evil Requiem's New Game Plus mode is a multi-step puzzle that ties together "The Girl" antagonist, the Severed Hand, Marie's Doll, and the Sun Quartz Box from Grace's chapters. Solving it unlocks a secret ending dialogue and special trophy. This guide covers every step and the RNA-to-symbol decoding.

SPOILER WARNING: This guide reveals key story content for the post-credits content. Do NOT read until you have completed the main story at least once.

Spoken by The Girl: "Let's play. GGC-AAG-AUA-ACG-UGU-CAU"

Why this puzzle exists

"The Girl" is a recurring antagonist throughout Grace's chapters — a child-like figure with bandaged hands who appears in dreams, hallucinations, and key story moments. After the main credits roll on a first playthrough, players who pay attention to her dialogue notice a strange chant: "Let's play. GGC-AAG-AUA-ACG-UGU-CAU."

This is RNA notation — the four letters G/A/U/C represent the four ribonucleotides (Guanine, Adenine, Uracil, Cytosine). Combined with the Severed Hand item Grace can find in NG+, these letters decode to a Sun-Moon-Star symbol sequence that opens a hidden puzzle in the Lead Researcher's Office.

Letter-to-symbol mapping

The four RNA letters correspond to celestial symbols based on clues hidden throughout the game:

G
☀️ Sun
// GUANINE
A
🌙 Moon
// ADENINE
U
⭐ Star
// URACIL
C
☀️ Sun
// CYTOSINE = SUN (PAIRED)
💡 How to discover this mapping: Three Files in the Care Center reveal the encoding through ARK research notes. They describe Cytosine and Guanine as "the bonded pair" — both mapped to Sun. Adenine pairs with Uracil — Moon and Star respectively. Reading all three Files unlocks an in-game File entry that confirms the mapping.

Decoded sequence

Applying the mapping to The Girl's chant:

// FINAL PUZZLE INPUT
Sun · Sun · Star · Sun · Moon · Star · Sun · Sun · Moon · Sun · Star · Moon · Star · Star · Sun · Sun · Moon · Star

The complete 18-symbol sequence breaks into 6 sets of 3, mirroring the original RNA codons:

  1. GGC → Sun · Sun · Sun
  2. AAG → Moon · Moon · Sun
  3. AUA → Moon · Star · Moon
  4. ACG → Moon · Sun · Sun
  5. UGU → Star · Sun · Star
  6. CAU → Sun · Moon · Star

Complete walkthrough

// PRE-NG+ PREPARATION (first playthrough)

// STEP 01
Find Marie's Doll in the Collection & Testing room (Care Center Basement). It looks like an old porcelain doll with bandaged hands — visually similar to The Girl.
// STEP 02
Store Marie's Doll in Grace's item box immediately. This is critical — items in the box carry over to NG+.
// STEP 03
Find the Severed Hand in the Care Center West Wing 2F (sealed locker, Level 2 Wristband required). Use the Laser Microscope in the Blood Lab to analyze it. Records the RNA sequence for NG+.
// STEP 04
Read all 3 RNA decoding Files: "Anomalous Cellular Replication" (Lab Notes A-12), "The G-C Bond" (Researcher's Diary 4), and "Bandaged Anomaly" (East Wing Patient File 11). These unlock the letter mapping.
// STEP 05
Complete the main story to credits. Save your clear data when prompted. NG+ unlocks.

// NG+ EXECUTION

// STEP 06
Start NG+. Marie's Doll appears in your starting inventory automatically.
// STEP 07
Progress through the story until you rescue Emily from the Care Center Basement.
// STEP 08
Return to the Blood Lab with Emily. Re-analyze the Severed Hand on the Laser Microscope. The Sun Puzzle in the Lead Researcher's Office becomes interactable again.
// STEP 09
Bring Emily AND Marie's Doll to the Lead Researcher's Office. Approach the Sun Puzzle Box. The puzzle now accepts 18 symbol inputs instead of 4.
// STEP 10
Input the decoded RNA sequence: Sun-Sun-Sun → Moon-Moon-Sun → Moon-Star-Moon → Moon-Sun-Sun → Star-Sun-Star → Sun-Moon-Star
// STEP 11
A hidden compartment opens. Inside is a special letter from Marie addressed to "The Girl." Picking it up triggers a unique cutscene.

What the puzzle reveals

The cutscene reveals Marie's connection to "The Girl" — they are sisters separated by Capcom's bioweapons program. Marie's Doll was originally The Girl's, lost when she was taken by Umbrella researchers years before the events of RE9.

The puzzle's solution implies that Marie chose to remain in the Care Center as a "guardian" of The Girl's spirit, refusing to leave even when offered escape. This thread closes a major emotional arc that's only hinted at in the main story.

Why this is the "best" RE9 secret

Capcom designed this puzzle as a true Easter egg for completionists. It requires:

It's the kind of puzzle that took the speedrun and 100% communities about two weeks after launch to fully decode and document. Now that the mapping is public, you can solve it on your second NG+ run.
